One way to do it is to make an items wanted list. On the list you can put a minimum quantity for each part on the list and then look to see stores that have the most items meeting list criteria (either by lot, unique lots, or total parts)

Find your part, in the color you want, and then click the link to the "lots for sale" page. When that page opens, pay special attention to the "Search Within Results" bar: At its rightmost side is a "sort by" field, which by default is usually set to the "Best Price" specification. Change this using the drop-down to "Highest Qty" instead. Then, hit "Go"...

That should give you what you need.

From there, I usually just browse until I find that seller with the most of what I need, at the most reasonable price. In reality, this often involves at least 2-3 separate sellers. A seller with hundreds of a certain kind of part in a certain kind of color, in my own experience, will also have a selection of hundreds of others in sister parts/ colors as well.

I generally though, begin searching with my most expectedly expensive or hardest-to-find part first, or those in which I need the most quantity.

Very hands on, sometimes time consuming, but its always worked for me.

There is another trick. If you build a wanted list of all the parts you need in what quantities, then go to your Wanted tab and select Wanted by Shop. In the sub selection choose by lots. This will then let you find sellers who have a large part of what you want all under one roof. Doing it this way can take 50 seperate orders and cut it down to 2 or 3. It is also worth playing a bit with the search options.

One word of warning. Be very very cafefiul with the auto fill function once you find a seller. It can be weird. Many sellers have multiple small lots of the same part on their stores. Typically from when they tell the system what set they have and let it auto part it out. The system does not always auto consolidate these. So if you need 10 1x2 bricks and a seller has 3 entries or lots of them on his store. Say of 25, 30 and 12. Auto fill will auto fill all 3 lots for 10 so you end up with 30 parts. You need to go back and clean it up and delete the unwanted ones.

One more thing to add: If you go to 'My BrickLink' tab, on the left there is a category labeled 'Search', below which is an option for 'My Settings', under which you can check the bottom box to allow filter options. These will show up at the top of each page when searching specific parts, right above the first part.

Using the filter options, you can still search by best price to have the cheapest first, but can also search only for stores with a minimum quantity (set by you) of the desired part.

You can also filter by country and, as notaromantic said, can also search by 'highest quantity' (seller with the most first) rather than the default 'best price' (seller with the cheapest first).

These filters help very much in speeding up BrickLink searching. Hope this helps!
